Comparative Histochemical Study on the Tracheal Cartilage



Abstract
A comparative histochemical study was done on the histochemical nature of mucosubstance and distribution of RNA and alkaline phosphatase in the matrix of the tracheal cartilage, using the human fetus, rat, guinea pig, rabbit, cat, dog and fowl.
The results were as follows:
1) The lacunar capsule of the tracheal cartilage were stained strongly positive with alcian blue, but not reacted with PAS, and strongly positive only to alcian blue with combined staining of alcian blue and PAS in the human fetus and all animal materials. RNA was moderately to strongly positive in the human fetus and all animal materials but alkaline phosphatase reaction was moderately positive in the rat and cat, and weakly positive in the other animal materials and human fetus.
2) The territorial matrix of the tracheal cartilage were stained moderately positive in the rat and fowl, strongly positive in the other animal materials and human fetus with alcian blue, but almost negative in the human fetus and all animal materials with PAS, and combined staining of alcian blue and PAS was moderately to strongly positive only to alcian blue in the human fetus and all animal materials. RNA was weakly to moderately positive in the human fetus and all animal materials, but alkaline phosphatase reaction was weakly to moderately positive in the rat and rabbit, but almost negative in the other animal materials and human fetus.
3) The granular matrix of the interterritorial matrix was stained weakly positive in the rat and fowl, moderately to strongly positive in the other animal materials and human fetus with alcian blue, but almost negative in the human fetus, rat and cat, and moderately to strongly positive in the other animal materials with PAS. Combined staining of alcian blue and PAS was strongly positive only to alcian blue in the human fetus, dog and cat, but moderately positive stronger to alcian blue in the rat, guinea pig and rabbit, and stronger to PAS in the fowl. RNA was moderately positive in the guinea pig, cat and dog, but weakly positive in the other animal materials and human fetus, and alkaline phosphatase reaction was weakly positive in the rat, but almost negative in the other animal materials and human fetus.
4) The granular matrix of the interterritorial matrix was stained weakly to moderately positive in the dog and human fetus, but almost negative in the other animal materials with alcian blue and almost negative in the human fetus and cat, but moderately to strongly positive in the all other animal materials with PAS. Combined staining of alcian blue and PAS was strongly positive only to alcian blue in the human fetus, but moderately to strongly positive only to PAS or stronger to PAS in the all animal materials. RNA was moderately positive in the guinea pig and dog, but weakly positive in other animal materials and human fetus, and alkaline phosphatase reaction was moderately positive in the rat, but almost negative in the other animal materials and human fetus,
5) The subperichondrial matrix of the tracheal cartilage was stained moderately positive in the human fetus and dog, but not stained in the other animal materials with alcian blue, and moderately positive in the human fetus and cat, but strongly positive in the other animal materials with PAS. Combined staining of alcian blue and PAS was strongly positive only to alcian blue in the human fetus, but strongly positive only to PAS or stronger to PAS in the all animal materials. RNA was moderately to strongly positive in the human fetus and all animal materials, but alkaline phosphatase reaction was moderately positive in the rat, guinea pig and rabbit, and weakly positive in the other animal materials and human fetus.
